Divers travaux menés depuis 1984 montrent qu'il existe, chez l'huître creuse Crassostrea gigas, des cellules montrant un nombre anormal de chromosomes (2n = 19, 18 ou même 17 au lieu de 2n = 20). Le niveau d'aneuploïdie est déterminé par le décompte des chromosomes à partir de suspensions cellulaires de tissu branchial. Le pourcentage de cellules aneuploïdes est toujours significativement supérieur dans les "lots de queue", c'est-à-dire les huîtres présentant des croissances plus faibles, et peut atteindre plus de 30 %. Récemment, une étude réalisée au sein du programme européen « Genephys » a démontré que plus de 50 % de la variance pour la vitesse de croissance était liée au taux d'aneuploïdie. L'étude de la variabilité génétique de l'huître creuse Crassostrea gigas est un point essentiel dans la mise en place de programmes de gestion des ressources génétiques et de sélection chez cette espèce. C. gigas, originaire du Japon, a été introduite dans de nombreuses régions du monde et représente aujourd'hui plus de 95 'ro de la production mondiale d'huîtres. A European programme (GENEPHYS) was launched in 1996 to study the genetic basis and variability of physiological characters involved in growth in Crassostrea gigas. This research programme was a prerequisite for any genetic selection programme on physiological functions. Research groups from Ireland, England, Greece and France are involved in this project and participate in genetic analyses (allozymes and microsatellites) and physiological analyses (ecophysiology, enzymology, stress proteins). Ifremer at La Tremblade was responsible for producing the biological material while Ifremer-CREMA (human-influenced marine ecosystems research centre) was responsible for carrying out non-destructive physiological analyses. |

The European flat oyster, Ostrea edulis, a native of Europe, occurs naturally from Norway to Morocco in the North-Eastern Atlantic and in the whole Mediterranean Basin; It has been a harvested species for at least 6000 years. Natural populations are also observed in eastern North America, from Maine to Rhode Island, following intentional introductions in the 1940s and 1950s. The species was also introduced in Canada for aquaculture purpose 30 years ago and some populations naturalised in Nova Scotia, New Brunswick and British Columbia. These stocks were imported from naturalised populations in Maine whose ancestors originated in the Netherlands. |

Crassostrea angulata and Crassostrea gigas, two cupped oyster taxa of Asian origin (O'Foighil et al, 1998), were successively introduced into Europe during the XVIth and the XXth century respectively (Héral and Grizel, 1991). To date, populations presumed to be C. angulata remain only in the south of Spain and Portugal, while the geographic range of C. gigas is expending, notably due to aquaculture. The two taxa can be distinguished by RFLP analysis of the mitochondrial Cytochrome Oxydase I locus (COI). The Pacific oyster, Crassostrea gigas (Thunberg), and the Portuguese oyster, C. angulata (Lamarck), are two closely related taxa. Although these two taxa were both introduced from Asia into Europe. one (C. gigas) was voluntarily introduced in the early 1970s, whereas the other (C. angulata) was presumed to be present in Europe for at least four centuries, but nearly disappeared because of disease. The G-banding technique was performed on aneuploid karyotypes from gill tissue of the Pacific oyster, Crassostrea gigas, to assess whether chromosome losses could be explained by differential chromosomal susceptibility and to clarify the negative correlation between aneuploidy and growth rate previously reported in different populations of this oyster. The study of 95 G-banded aneuploid karyotypes showed that only 4 of the 10 chromosome pairs (viz. 1, 5, 9, and 10) of C. gigas were affected by the loss of one homologous chromosome. Pairs 1, 9, and 10, which were lost in 56, 33, and 44% of cases, respectively, may be considered to be differentially affected. Quantification of mRNA of genes related to metabolism, immunity and cellular stress was examined in relation to a massive mortality event during the culture of American oyster larvae, Crassostrea virginica which was probably, in regard to previous microbiological analysis, induced by vibrio infection. Mortalities of oyster Crassostrea gigas seed associated with ostreid herpesvirus OsHV-1 μVar have been observed in many oyster-producing countries since 2008. The present study, comprised of 4 complementary experiments, aimed to identify factors associated with disease-induced oyster mortality in order to propose mitigation strategies. Our first experiment compared survival of oysters from natural spatfall with others sampled from nurseries, after thermal elevation in the laboratory from <14 to 21°C.
